INTRODUCTION:
hytic&nbs;acid&nbs;(hytate;&nbs;myo-inositol&nbs;1,2,3,4,5,6,&nbs;hexakishoshate)&nbs;is&nbs;the
rimary&nbs;source&nbs;of&nbs;inositol&nbs;and&nbs;storage&nbs;hoshorus&nbs;in&nbs;lant&nbs;seeds
contributing&nbs;~70&nbs;%&nbs;of&nbs;total&nbs;hoshorus.&nbs;The&nbs;abundance&nbs;of&nbs;hytic&nbs;acid
in&nbs;cereal&nbs;ains&nbs;is&nbs;a&nbs;concern&nbs;in&nbs;the&nbs;foods&nbs;and&nbs;animal&nbs;feeds&nbs;industries
because&nbs;the&nbs;hoshorus&nbs;in&nbs;this&nbs;form&nbs;is&nbs;unavailable&nbs;to&nbs;monogastric
animals&nbs;due&nbs;to&nbs;a&nbs;lack&nbs;of&nbs;endogenous&nbs;hytases;&nbs;enzymes&nbs;secific&nbs;for
the&nbs;dehoshorylation&nbs;of&nbs;hytic&nbs;acid.&nbs;In&nbs;addition,&nbs;the&nbs;strong&nbs;chelating
characteristic&nbs;of&nbs;hytic&nbs;acid&nbs;reduces&nbs;the&nbs;bioavailability&nbs;of&nbs;other
essential&nbs;dietary&nbs;nutrients&nbs;such&nbs;as&nbs;minerals&nbs;(e.g.&nbs;Ca2+,&nbs;Zn2+,&nbs;Mg2+,
Mn2+,&nbs;Fe2+/3+),&nbs;roteins&nbs;and&nbs;amino&nbs;acids.1&nbs;High&nbs;hytic&nbs;acid&nbs;content
feeds&nbs;are&nbs;generally&nbs;sulemented&nbs;with&nbs;inorganic&nbs;hoshate,&nbs;however
this&nbs;causes&nbs;increased&nbs;faecal&nbs;hoshate&nbs;levels&nbs;and&nbs;subsequent
eutrohication&nbs;of&nbs;waterways.&nbs;Alternatively,&nbs;sulementation&nbs;with
mercial&nbs;hytases&nbs;is&nbs;being&nbs;increasingly&nbs;oular&nbs;and&nbs;reduces&nbs;the
requirement&nbs;for&nbs;inorganic&nbs;hoshate&nbs;sulementation&nbs;as&nbs;well&nbs;as&nbs;the
associated&nbs;environmental&nbs;issues.
Currently,&nbs;there&nbs;is&nbs;no&nbs;mercially&nbs;available,&nbs;simle,&nbs;quantitative
method&nbs;for&nbs;hytic&nbs;acid&nbs;and&nbs;while&nbs;such&nbs;measurement&nbs;is&nbs;relatively
lex&nbs;the&nbs;generally&nbs;acceted&nbs;AOAC&nbs;Method&nbs;986.11&nbs;has
limitations.2&nbs;For&nbs;each&nbs;individual&nbs;analysis&nbs;the&nbs;method&nbs;requires
cumbersome&nbs;anion-exchange&nbs;urification&nbs;and&nbs;a&nbs;major&nbs;inherent
assumtion&nbs;here&nbs;is&nbs;that&nbs;only&nbs;hytic&nbs;acid&nbs;is&nbs;urified.&nbs;While&nbs;this
assumtion&nbs;is&nbs;viable&nbs;for&nbs;non-rocessed&nbs;ains&nbs;for&nbs;which&nbs;hytic&nbs;acid
rises&nbs;at&nbs;least&nbs;97&nbs;%&nbs;of&nbs;total&nbs;inositol&nbs;hoshates,&nbs;it&nbs;is&nbs;not&nbs;viable&nbs;for
rocessed&nbs;foods&nbs;and&nbs;feeds&nbs;which&nbs;can&nbs;contain&nbs;higher&nbs;levels&nbs;of&nbs;some
lower&nbs;myo-inositol&nbs;hoshate&nbs;forms&nbs;(i.e.&nbs;I3,&nbs;I4,&nbs;I5)&nbs;that&nbs;would&nbs;coelute
with&nbs;hytic&nbs;acid&nbs;and&nbs;contribute&nbs;to&nbs;overestimation&nbs;of&nbs;the&nbs;hytic
acid&nbs;content.3,4
Given&nbs;the&nbs;lexities&nbs;of&nbs;the&nbs;urification&nbs;and&nbs;measurement&nbs;of&nbs;hytic
acid&nbs;searate&nbs;from&nbs;lower&nbs;myo-inositol&nbs;hoshate&nbs;forms,&nbs;Megazyme
has&nbs;develoed&nbs;a&nbs;simle,&nbs;quantitative&nbs;method&nbs;(K-HYT)&nbs;to&nbs;measure
total&nbs;“available&nbs;hoshorus”&nbs;released&nbs;from&nbs;food&nbs;and&nbs;feed&nbs;samles&nbs;that
is&nbs;amenable&nbs;to&nbs;high&nbs;numbers&nbs;of&nbs;samles&nbs;and&nbs;does&nbs;not&nbs;require&nbs;tedious
anion-exchange&nbs;urification.&nbs;This&nbs;method&nbs;involves&nbs;acid&nbs;extraction&nbs;of
inositol&nbs;hoshates&nbs;followed&nbs;by&nbs;treatment&nbs;with&nbs;a&nbs;hytase&nbs;that&nbs;is
secific&nbs;for&nbs;hytic&nbs;acid&nbs;(I6)&nbs;and&nbs;the&nbs;lower&nbs;myo-inositol&nbs;hoshate
forms&nbs;(i.e.&nbs;I2,&nbs;I3,&nbs;I4,&nbs;I5).&nbs;Subsequent&nbs;treatment&nbs;with&nbs;alkaline
hoshatase&nbs;ensures&nbs;the&nbs;release&nbs;of&nbs;the&nbs;final&nbs;hoshate&nbs;from&nbs;myoinositol
hoshate&nbs;(I1)&nbs;which&nbs;is&nbs;relatively&nbs;resistant&nbs;to&nbs;the&nbs;action&nbs;of
hytase.&nbs;The&nbs;total&nbs;hoshate&nbs;released&nbs;is&nbs;measured&nbs;using&nbs;a&nbs;modified
colorimetric&nbs;method&nbs;and&nbs;given&nbs;as&nbs;ams&nbs;of&nbs;hoshorus&nbs;er&nbs;100&nbs;g&nbs;of
samle&nbs;material.5,6

Alkaline&nbs;hoshatase&nbs;will&nbs;also&nbs;release&nbs;hoshate&nbs;from
monohoshate&nbs;esters&nbs;other&nbs;than&nbs;myo-inositol&nbs;hoshate,&nbs;however
analyses&nbs;of&nbs;a&nbs;broad&nbs;sectrum&nbs;of&nbs;samles&nbs;using&nbs;K-HYT&nbs;indicated&nbs;that
hoshorus&nbs;released&nbs;from&nbs;monohoshate&nbs;esters&nbs;other&nbs;than&nbs;myoinositol
hoshate&nbs;is&nbs;small.
